【摘要】:ThisguideisintendedforuseinanylaboratoryutilizingPCRorRT-PCRtoamplifyanddetectnucleicacidsequencesofmycobacteriafromabiologicalpreparationandtoidentifythespeciesoforigin.ThecriteriausedfortheidentificationandevaluationoftheamplificationreactionsshouldbeadministeredbyanindividualtrainedintheuseofmolecularbiologicalandmicrobiologicaltechniquesassociatedwithPCRandMTB.1.1Thisguidecoversbasicconsiderations,criteria,principlesandrecommendationsthatshouldbehelpfulwhendeveloping,utilizing,orassessingPCR-specificprotocolsfortheamplificationanddetectionoridentificationofmycobacterialnucleicacids.Thisguideisnotaspecificprotocolforthedetectionofspecificmycobacteria.Itisintendedtoprovideinformationthatwillassisttheuserinobtaininghighqualityandreliabledata.TheguideiscloselyrelatedtoandshouldbeusedconcurrentlywiththegeneralPCRGuideE1873.1.2Thisguidehasbeendevelopedforuseinanymolecularbiologyorbiotechnologylaboratory.Itmaybeusefulforthedetectionofmycobacteriainclinical,diagnosticlaboratories.1.3ThisguidedoesnotcoverdetailsofthevariousmethodssuchasgelelectrophoresisthatcanbeutilizedtohelpidentifyPCR-amplifiedmycobacterialnucleicacidsequences,anditdoesnotcoverdetailsofinstrumentcalibration.1.4ThisguidedoesnotcoverspecificvariationsofthebasicPCRorRT-PCRtechnology(forexample,quantitativePCR,multiplexPCRandinsituPCR),anditdoesnotcoverdetailsofinstrumentcalibration.1.5Warning-Laboratoryworkinvolvingcertainclinicalspecimensandmicroorganismscanbehazardoustopersonnel.Precaution:BiosafetyLevel2facilitiesarerecommendedforpotentiallyhazardouswork,andBiosafetyLevel3facilitiesarerequiredforpropagatingandmanipulatingMycobacteriatuberculosiscultures().SafetyguidelinesshouldbeadheredtoaccordingtoNCCLSM29-T2,I17-Pandotherrecommendations().
